Proiektuaren lehen faseko azken saioak! Hurrengo hilabetean pipeteoaren, geneen, bakterioen eta lehen eskuko landareen atalera joango ginen, eta horretarako ahalik eta informazio argiena izan behar genuen, eta horretan aritu ginen!

Kontutan hartzeko punturik garrantzitsuenak hauek izan ziren:

Zer da, eta nola funtzionatzen du plasmido batek?

Plasmidoa DNA molekula zirkular bat da. Biologia sintetikoan erabiltzen da, geneetan dauden intereseko ezaugarri berriak informazio-bektoreak diren bakterioei eta erreakzioen azken produktuei transmititzeko.

Nola sartzen da bakterio baten barruan sortu dugun plasmidoa?

Elektroporazioaren metodoa erabiltzen da, produktu aktiboak irrati-maiztasuneko eta goi-tentsioko eremu elektromagnetiko baten bidez sartzen laguntzen duen teknika.

Zer da miniprep teknika?

Minipreparen teknika bakterio-hazkuntza batetik plasmako DNA ateratzea da.

Plasmidoak atera ondoren, espektrofotometroaren bidez neurtzen da plasmidoen kontzentrazioa. Espektrofotometroak, gainera, arazo ugari sortuko lituzke aurrerago, eta hainbat nahaste eragingo lituzke (bitxia da kontzentrazio batek balio negatiboa izatea, ezta?).

Nola jakin dezakegu plasmidoak bakterioan daudela?

Horretarako, plasmidoak zenbait entzima erabiliz “mozten” ditugu, eta, ondoren, neurketa bat egiten dugu, 20 eta 1.000 base-pare bitarteko DNA zati guztiak ikusteko, gure geneak BioBrick-etan ondo txertatuta zeuden ikusteko.

Informazio-ponpa horren guztiaren ondoren, gure batak estreinatu nahi genituela esatea besterik ez zaigu geratzen. Azkenik!

March: Last sessions before de lab

Last sessions of the first phase of the project! The following month we would move on to handling first hand pipetting, genes, bacteria and plants, and for that we had to leave the Planetarium with what we learned as clearly as possible, and that is what we were dedicating ourselves to!

The most important comprehension points were the following:

What is a plasmid and how does it work?

A plasmid is a circular DNA molecule, which is used in synthetic biology to transmit the new characteristics of interest, found in genes, to bacteria that are both information vectors and the end products of reactions.

How does the plasmid that we have created get into bacteria?

The electroporation method is used, it is a technique that favors the penetration of active products through the action of a high voltage radio frequency electromagnetic field.

What is the Miniprep technique?

The Miniprep technique is the extraction of plasmid DNA from a bacterial culture.

Once the plasmids have been extracted, the concentration of these is measured with a spectrophotometer, a device that would certainly give us quite a few problems later on and would cause several confusions (it is a bit strange that a concentration comes out with a negative value, right?)

How can we know that the plasmids are actually found in the bacteria?

To do this, we "cut" the plasmids using different enzymes, and then we make a measurement with which we can see all the fragments that have between 20 and 1000 base pairs, to be able to see if our genes were really well inserted in the BioBricks.

After all this burst of information, we can only say that we were looking forward to finally using our lab coats!
